Virtual Dilution Built-in

Fast and easy spectrophotometric quantification

The OD600™ has been especially designed for the spectrophotometrical analysis of bacterial and yeast growth rates and the quantification of proteins at a wavelength nearby 600 nm (e.g. Bradford). Combining OD600™ with Implen’s DiluCell™ 10 or DiluCell™ 20 facilitates measurements with low sample volumes of 200 µl and 100 µl, respectively and a virtual sample dilution of either factor 10 or 20. The instrument is battery operated and can be easily transported for usage in incubation cabinets and under anaerobic conditions. The batteries are recharged automatically by connecting to the mains and last for almost 1 month when fully charged. A 600nm LED source in combination with a fibre optic is used to obtain the optical density measurement. Spillages can be easily wiped from the even surface, and the cell compartment area can be rinsed with, formaldehyde or ethylene oxide to sterilize it.

Monitoring virtual dilution with DiluCell™

The same red-coloured sample was filled in the two different available DiluCell™ types DC 20 (left cell) and DC 10 (right cell), respectively. The automatic virtual dilution can be seen by the weaker red colour within the DC 20 generating a 1/20 dilution. The use of this new disposable cuvette allows the lowering of the sample volume to 100 µl or 200 µl, respectively. The automatic dilution of the sample combined with the added benefit of low sample volume make the DiluCell™ essential in every modern laboratory.

Measuring Cell Density

The stage of growth of a bacterial culture needs to be monitored to ensure that the cells are harvested at the optimum point for the greatest density of live cells. An exemplary growth curve is given. Cells should be harvested towards the end of the log phase. The optical density of the sample indicates when this point has been reached. This value varies dependent on the cells being grown. Routinely the cells are grown until the absorbance at 600 nm (known as OD 600) reaches approximately 0.4 prior to induction or harvesting. A linear relationship exists between cell number (density) and OD 600 up to approx. 0.6. Combining OD600™ with Implen’s DiluCell™ 10 or DiluCell™ 20 allows the quantification of bacterialand yeast cultures with low sample volumes of 200 µl and 100 µl, respectively without prior dilution. The cultures are virtually diluted with either factor 10 or 20 and the measurements can be taken directly at the laboratory area where cells are cultured.

Protein Quantification with Bradford Assay

Bradford Assay is a rapid and accurate method used to determine the total protein concentration of a sample by quantifying the binding of a dye, Coomassie Brilliant Blue G-250, to an unknown protein.

The assay is based on the observation that the absorbance maximum for an acidic solution of Coomassie Brilliant Blue G-250 shifts from 465 nm to 595 nm when binding to protein occurs.
By comparing this binding to that of different, known concentrations of a standard protein, the concentration of the unknown protein can be calculated. Standard curves with bovine serum albumin (BSA) are given as an example.

The OD600™ a is battery powered system with a built in memory to store up to 99 readings. Data can be then printed out on the OD600™ thermal paper printer (optional) or uploaded on a computer via an RS232 interface. The OD600™ is configured to work with DiluCell™ but at the same time can take standard 10 mm glass or plastic cells (macro and semi micro).
